Bio
Pulmonary alveolar Type II cells are vital for normal lung function since they synthesize and secrete surfactant and serve as progenitor cells for the alveolus following lung injury. The laboratory has recently developed methodology to transfect isolated Type II cells utilizing HVJ envelopes, allowing introduction of siRNA and cDNA into primary cultures of Type II cells. This methodology will now be utilized to explore factors regulating secretion of lamellar bodies, and more specifically, secretion of Surfactant Protein C.
The methodology utilized in the laboratory includes generation of multiple transgenic mouse lines, techniques for molecular morphology, tissue culture, cell preparation, and lung physiology as well as standard techniques of molecular biology including Northern, Western, Southern blotting, RT-PCR, cloning and library screening.
