Movie 1. PGCs at E9.0Movie 1. PGCs at E9.0. The trunk region of an E9.0 Oct4?PE:GFP embryo was dissected, cultured and filmed for 12 hrs. PGCs (green) can be seen moving around and between the cells of the hindgut. At this stage they are confined within the hindgut and do not exhibit directional movement with respect to the A/P or D/V axes.
Movie 2. PGCs at E9.5Movie 2. PGCs at E9.5. A transverse slice was cut from the trunk of an E9.5 Oct4?PE:GFP embryo, cultured and filmed for 12 hrs. PGCs emerge from the dorsal most aspect of the hindgut, split into two streams and migrate towards the positions of the genital ridges.
Movie 3. PGCs at E10.5Movie 3. PGCs at E10.5. A transverse slice was cut from the trunk of an E10.5 Oct4?PE:GFP embryo, cultured and filmed for 12 hrs. PGCs continue to migrate towards the positions of the genital ridges. At this stage they are slowing down and beginning to form clusters. Most germ cells left on the midline and in the hindgut mesentery fragment and die.
