• Mouse Cytogenetics Core

    Chromosome karyotyping and molecular cytogenetics analyses play an important role in the study of chromosome abnormalities, genetic aberrations, cell line characterization and establishing disease models.

    The Mouse Cytogenetics Core at the Cincinnati Children’s Division of Human Genetics offers research investigators support for mouse classic karyotyping and molecular cytogenetic studies. Services include chromosome analysis of mouse tissues and cell lines, karyotype characterization of embryonic stem cells and induced pluripotent stem cells (iPSC), and locus copy number enumeration, refining chromosome breakage sites, and mapping of transgene integration sites.

    *Each BAC / PAC DNA preparations may yield 10-15 ug DNA.
    **Each labeling reaction may be good for 7-10 FISH assays.

    Mouse Stem Cells (ES or iPS)

    A minimum of two wells on a 24-well plate per clone, and it is preferable to have 3-4 wells plated at varying densities. The clones need to be 50-80% confluent and require media change the night before submission.

    Mouse Tissue (Fibroblasts)

    Fresh skin (ear, tail), fresh lung, or embryo (mouse embryonic fibroblasts)

    Minimum sample sizes: 6 mm ear, 1 cm tail clip, ½ lobe of lung, early passage fibroblast culture.

    Tissue is collected and sent to the laboratory in sterile transport media at room temperature. Sample must be received with a completed requisition form. Tissues can be stored (refrigerated in transport media for 24 hours), but should be set up fresh when possible. Transport media available upon request.
