Flow Cytometry and Cell Sorting Instrumentation

The flow cytometry and cell sorting facility at Cincinnati Children's Hospital Medical Center is equipped with two FACSCaliburs, two FACSVantage SE high speed cell sorters and a FACSCanto system, located in room 1315 of the old research building.

FACSCalibur Analyzers (Two on Location)

4 color and 8 parameters (light scatter, 4 colors, pulse processing, and time) with Autosampler for 12x75 test tubes.

Two lasers:

  • 1 air-cooled Argon laser (488 nm)
  • 1 air-cooled red diode laser (635nm)

Interfaced via Macintosh G4 with CellQuest Pro software.

FACSCanto Analyzer

FACSCanto Analyzer Customized for 10 parameters (8 colors, forward scatter and side scatter)

Two lasers:

  • 498 air-cooled 20 mW solid state
  • 633 nm 77 mW HeNe

FACSVantage SE Cell Sorters (Two on Location)

Both instruments house three lasers (488 nm, 633 nm, and UV), advanced optics and provide enormous flexibility in terms of applications as well as unprecedented sensitivity. High speed multi-laser sorts are performed routinely with throughputs near 20,000 cells/sec, < 5% aborts and majority of sorted fractions exhibiting > 98.5% purity.

The FACSVantage is equipped with hi-speed digital sorting electronics, 4 way simultaneous sorting, 8 PMT's, forward scatter and side scatter. The CloneCyt Plus (ACDU Sort) option sorts cells onto glass slides or into 96 well microtiter trays.
