Single Cell Gene Expression Assays – Fluidigm C1
Recently the core has started using a new service that generates RNA sequencing data from single cells. Core personal use a Fluidigm C1™ Single-Cell Auto Prep System running a protocol designed to capture and isolate single cells that are subsequently lysed. The RNA from each cell is then converted into full-length cDNA that is amplified by PCR. The core then uses an assay from Illumina™ to generate libraries for each cell. The libraries are pooled and submitted to the DNA Sequencing and Genotyping Core for sequencing.
