Ectoderm Differentiation
Neuroepithelia
A fibroblast-derived, integration-free iPSC line generated using episomal plasmids was differentiated to neuroepithelia by exposure to 10uM SB431542 and 2uM dorsomorphin for 10 days. Efficient generation of neuroepithelia was confirmed by immunofluorescence detection of Pax6 expression. Neural progenitors formed using this method have been reported to form neural rosettes and be amenable to directed differentiation into a variety of neural cell types.
Neural Stem Cells
H9 hESCs were differentiated to neural progenitors using STEMdiff™ Neural Induction Medium. Generation of neural progenitors was confirmed by formation of neural tube-like rosette structures and by immunofluorescence detection of Nestin and Sox1 expression. Neural rosettes are believed to be capable of differentiating into all neural cell types.
